چکیده

The use of “Housekeeping proteins” as loading controls in western blots requires that they do not change their level of expression. However, deviations from this rule have been observed. This study investigated the expression of the commonly used proteins HSP60, -tubulin, -actin and GAPDH by western blot during retinoic acid-induced differentiation of the embryonal carcinoma NTera-2 and the neuroblastoma SH-SY5Y cell lines. -actin was found to be a poor loading control, increasing significantly during differentiation, whereas HSP60 and GAPDH were the more evenly expressed of those analysed. Our results underline the importance of selecting accurate loading controls in this biological

Induction of Human Embryonic Stem Cells into neuronal differentiation by increasing cyclic Adenosine Mono Phosphate

Introduction: To evaluate the cAMP -mediated IBMX (3-IsoButyle -1-Methyl Xanthin) and db-cAMP (dibutyryl cAMP) effects on differentiation of human Embryonic Stem Cells (hESCs) into nerve cells were the objectives of this study. Methods: We have used Royan H1 hESC- derived embryoid bodies with four treatment groups: six days treatment with IBMX (5×10 -4M) and db-cAMP (10 -9M) (referred to as...

Expression pattern of neurotrophins and their receptors during neuronal differentiation of adipose-derived stem cells in simulated microgravity condition

Objective(s): Studies have confirmed that microgravity, as a mechanical factor, influences both differentiation and function of mesenchymal stem cells. Here we investigated the effects of simulated microgravity on neural differentiation of human adipose-derived stem cells (ADSCs). Materials and Methods:We have used a fast rotating clinostat (clinorotation) to simulate microgravity condition. R...

Laminar Organization of Cerebral Cortex in Transforming Growth Factor Beta Mutant Mice

Transforming growth factor betas (TGF?s) are one of the most widespread and versatile cytokines. The three mammalian TGF? isoforms, ?1, ?2, and ?3, and their receptors regulate proliferation of neuronal precursors as well as survival and differentiation in neurons of developing and adult nervous system. Functions of TGF?s has a wide spectrum ranging from regulating cell proliferation and differ...

EF1α is a suitable housekeeping gene for RT-qPCR analysis during osteogenic differentiation of mouse bone marrow-derived mesenchymal stem cells.

The expression of predominant housekeeping genes used in RT-qPCR can vary during development and differentiation. The frequently used housekeeping genes (ACTB, GAPDH, 18S rRNA, EF1α and RPL 13a) were evaluated during an early stage of the osteogenic differentiation of mouse bone marrow-derived mesenchymal stem cells (mMSCs) (under normal conditions or treated with CCG-4986) to identify housekee...

Sensitivity of dopaminergic neuron differentiation from stem cells to chronic low-dose methylmercury exposure.

Perinatal exposure to low doses of methylmercury (MeHg) can cause adult neurological symptoms. Rather than leading to a net cell loss, the toxicant is assumed to alter the differentiation and neuronal functions such as catecholaminergic transmission. We used neuronally differentiating murine embryonic stem cells (mESC) to explore such subtle toxicity. The mixed neuronal cultures that formed wit...
